# FDA Updates Internal Procedures for Handling Supplements

FDA has updated an internal policy document outlining how CBER staff process supplements to approved drug and biologic applications, detailing administrative steps used during regulatory review.

The [document](https://www.fda.gov/media/108895/download?attachment&ref=fdaweb.com) — Standard Operating Policy and Procedure (SOPP) 8401.2 Administrative Processing of BLA and NDA Supplements — provides guidance for staff on the administrative handling of supplements and outlines how they should receive, classify, and review these submissions, including those subject to user-fee programs such as the Prescription Drug User Fee Act and the Biosimilar User Fee Act.

Under the procedures, review teams must conduct several structured checkpoints during the regulatory process. These include an initial committee meeting to establish the review schedule and determine whether the supplement qualifies for standard or priority review, as well as a filing decision meeting where the agency determines whether the submission contains enough information to proceed with a full review.

The SOP also describes the use of mid-cycle review meetings, during which FDA staff evaluate the progress of the review, identify major deficiencies or safety concerns, and determine whether additional information requests should be sent to the sponsor.

If reviewers determine that key data or documentation are missing, the agency may issue a “refuse-to-file” decision or send formal information requests asking companies to submit additional material to support their application.

According to the agency, these SOPPs are published publicly as part of its transparency efforts and to help industry understand internal review processes used by regulators.

The updated procedure also describes how review teams document regulatory decisions, handle amendments submitted during the review, and coordinate approval letters and post-approval commitments once a supplement is cleared.
