First Approval Under New Priority Voucher Program

FDA has approved Augmentin XR (amoxicillin–clavulanate potassium) through the new Commissioner’s National Priority Voucher (CNPV) pilot program, permitting new U.S.-based production capacity at a domestic facility. This is the first product approved under the initiative aimed at strengthening U.S. drug manufacturing and supply chain resilience. The agency has not identified the manufacturer.

The decision came just two months after the application was filed — a significantly shortened timeline for this type of review. FDA commissioner Marty Makary said the pilot program is designed to rebuild domestic production capacity after decades of reliance on overseas suppliers for key medicines.

“Over the last few decades, America lost control of supply chains for key medicines we depend on,” he said. “That chapter is over – we’re entering a new era of manufacturing here at home,” “This first drug approval under the CNPV pilot program will strengthen domestic manufacturing and increase our national security.”

The agency said the Augmentin XR application aligned with the program’s stated priorities by demonstrating new U.S.-based production capacity at a domestic facility. It said the approval is expected to help mitigate longstanding antibiotic shortages driven by global supply chain disruptions and spikes in clinical demand. Over the past 20 years, shortages of commonly used antibiotics — including amoxicillin and Augmentin XR — have resulted in treatment delays and forced clinicians to rely on broader-spectrum alternatives. FDA has logged seven shortage reports for amoxicillin and two for Augmentin XR in recent years.

FDA said a multidisciplinary review team handled the application, incorporating drug substance, drug product, manufacturing, facility, and biopharmaceutics expertise. It said integrated quality assessments and frequent communication with the sponsor enabled the two-month turnaround and met the program’s target review window.

Augmentin XR is an oral combination antibiotic containing amoxicillin and the β-lactamase inhibitor clavulanate. It is approved to treat community-acquired pneumonia and acute bacterial sinusitis in adults and pediatric patients.
