Genmab Files sBLA for Darzalex for Multiple Myeloma

Genmab has submitted a supplemental BLA for Darzalex (daratumumab) and its combination use with lenalidomide and dexamethasone, or bortezomib and dexamethasone, for treating patients with multiple myeloma who received at least one prior therapy. The company has asked for a priority review, and the agency granted a breakthrough therapy designation last month.

The submission includes data from two Phase 3 studies — the CASTOR study of daratumumab in combination with bortezomib and dexamethasone versus bortezomib and dexamethasone alone in patients with relapsed or refractory multiple myeloma, and the POLLUX study involving daratumumab in combination with lenalidomide and dexamethasone versus lenalidomide and dexamethasone alone in the same patient population.

Darzalex is currently approved for intravenous infusion for the treatment multiple myeloma patients who have received at least three prior lines of therapy, including a proteasome inhibitor (PI) and an immunomodulatory agent, or who are double-refractory to a PI and an immunomodulatory agent. Daratumumab is a human IgG1k monoclonal antibody that binds with high affinity to the CD38 molecule, which is highly expressed on the surface of multiple myeloma cells, according to the company.
