Rhythm Reports Positive Data on Obesity Drug
Rhythm Pharmaceuticals is announcing positive topline results from its Phase 2 trial evaluating bivamelagon (formerly LB54640) in patients with acquired hypothalamic obesity. The investigational oral melanocortin-4 receptor (MC4R) agonist achieved statistically significant and clinically meaningful reductions in body mass index at 14 weeks of treatment, the company says.
Rhythm says it plans to request an End-of-Phase 2 meeting with FDA to seek guidance on a planned Phase 3 trial. It is also refining the formulation of bivamelagon to improve tolerability before initiating a Phase 3 trial.
The company says acquired hypothalamic obesity is a rare form of obesity that occurs following damage to the hypothalamic region of the brain, which includes the melanocortin-4 receptor pathway that controls physiological functions such as hunger and weight regulation. “Acquired hypothalamic obesity most frequently follows the growth or surgical removal of craniopharyngioma, astrocytoma or other rare brain tumors,” Rhythm says. Additional adverse effects may include traumatic brain injury, stroke, or inflammation due to infection.
